ABSTRACT

Objective@#To explore the prevalence of sexting among secondary vocational school students and to examine the correlation between sexting and mental health, so as to provide evidence for prevention and intervention on adolescents mental health.@*Methods@#In April 2021, three secondary vocational schools from central urban area, outskirts and outer suburbs in Shanghai, among 1 580 students were selected by cluster sampling.@*Results@#The proportions of students who received and who forwarded others private photos or videos were 11.96% and 2.78%, respectively. The proportion of students who sent others personal photos or videos was 2.54%. And the proportions of boys were higher than those of girls ( χ 2=45.89, 24.09, 6.66, P <0.01). The results of multivariable Logistic analysis showed that compared with those who had never sexted, students who had ever had any sexting were at higher risk of anxiety ( OR=2.40, 95%CI =1.59-3.61), depression( OR=1.86, 95%CI =1.25-2.75), non suicidal self injury intention ( OR=2.34, 95%CI =1.48-3.69) and suicidal intention ( OR=2.96, 95%CI =1.83-4.79)( P < 0.05 ).@*Conclusion@#There is a significant correlation between sexting and mental health among secondary vocational school students. The latent risks of sexting should be included in the curriculum of comprehensive sex education for teenagers to enhance their media literacy and decision making capability, so as to prevent sexual assault or abuse caused by sexting and promote sexual and reproductive health and as well as mental health of adolescents.

ABSTRACT

Placental blood transfusion can be achieved by delayed cord clamping and umbilical cord milking.The World Health Organization has proposed delayed cord clamping as the standard care for newborns that do not require resuscitation.In recent years, umbilical cord milking has become an alternative to delayed cord clamping due to the short duration, quick recovery of the fetus after delivery, and similar effect on placental blood transfusion to that of the delayed cord clamping.This study aims to summarize the research results of the two intervention methods, and their potential benefits and risks, aiming to provide references for the optimal umbilical cord cutting.

ABSTRACT

Objective@#To understand the prevalence and influencing factors of Children s Dental Fear (CDF) among 12-15 year-old children in Shenzhen city, so as to provide a framework to alleviate CDF and promote oral health.@*Methods@#Multi-stage stratified random sampling was used to select 5 509 children, aged 12-15 years, to participate in this study, which was carried out from October to November, 2018. The Children s Fear Survey Scheduling-Dental Subscale (CFSS-DS) and oral health examinations were conducted in Shenzhen.@*Results@#The prevalence of dental fear in children aged 12, 13, 14, and 15 years in Shenzhen was 30.3%, 30.5%, 33.6%, and 26.9%, respectively(χ 2=11.97, P=0.01). The CFSS-DS scores were(29.86±13.23)(29.72±13.59)(31.23±14.47)(29.79±13.24), respectively(F=3.60, P=0.01). The CFSS-DS scores of male and female participants were (27.92±13.69) and (32.62±13.12)(t=-12.97, P<0.01), respectively, and the CDF prevalence rates were 23.8% and 38.8%, respectively(χ 2=159.29, P<0.01). Multivariate Logistic regression analysis showed that gender, brushing frequency, visiting experience, oral knowledge level, oral health attitude, oral health and general health status were correlated with CDF (P<0.05).@*Conclusion@#Attention should be paid to the prevalence of dental phobia among children aged 12-15 years in Shenzhen, and comprehensive intervention measures should be taken to improve children s oral health.

ABSTRACT

Objective To analyze the effect of trametes robiniophila on the apoptosis and the expressions of invasion and metastasis related matrix metalloproteinase 2 (MMP-2) and MMP-9 in human gastric carcinoma poorly differentiated cell line MKN-45 and medium differentiated cell line SGC-7901.Methods Human gastric carcinoma cell lines MKN-45 and SGC-7901 incubated with trametes robiniophila at the different concentrations [0 (the negative control group), 5, 10, and 20 mg/ml] for 24 h. When bifluorescently stained with acridine orange-ethidium bromide (AO-EB), morphology was observed by using microscopy. Flow cytometry was used to test the apoptosis ratio after 24, 48, and 72 h. Reverse transcription polymerase chain reaction (RT-PCR) was applied to analyze the expressions of mRNA of MMP-2 and MMP-9 after 24 h, and the absorbance values of the bands after gel electrophoresis were used as their expression levels. Results Under the fluorescence microscope, the cells of the negative control group were green (normal cells), and the membrane was even in size and integrity. The proportion of apoptotic and necrotic cells was increased with the concentration enrichment of trametes robiniophila. The apoptotic cells were yellow, and their cell membrane lost integrity. Apoptotic bodies were found in cancer cells, and cell membrane showed bud projection. The nuclei of the apoptotic cells showed brightly condensed chromatin or fragmented. Chromatin was strongly stained and located in karyotheca. The necrotic cells were dyed red with integrity of size. The apoptotic ratio of MKN-45 cell line induced by trametes robiniophila after 24 h was (6.5 ±0.8)%, (14.6±1.0)%, (18.0±1.1)%, and (23.1±1.2)%, respectively (F= 333.972, P< 0.01) in negative control group and 5, 10, and 20 mg/ml trametes robiniophila group. After 48 h, the apoptotic ratio was (7.3 ±1.2)%, (18.3 ± 1.6)%, (24.5±1.3)%, and (27.2±1.7)%, respectively (F= 528.432, P= 0.001); after 72 h, the apoptotic ratio was (7.5 ±0.9)%, (50.2 ±1.6)%, (58.0 ±1.9)%, and (69.0 ±1.4)%, respectively (F= 3814.238, P< 0.01). After SGC-7901 cell line induced by trametes robiniophila for 24 h, the apoptotic ratio was (12.9 ±1.0)%, (19.4 ± 1.2)%, (22.0±1.7)%, and (23.0±1.9)%, respectively (F= 120.190, P< 0.01) in negative control group and 5, 10, and 20 mg/ml trametes robiniophila group. For 48 h, the apoptotic ratio was (10.2 ±1.3)%, (40.9 ±1.4)%, (51.6 ±1.9)%, and (66.2 ±1.9)%, respectively (F= 1281.342, P< 0.01). For 72 h, the apoptotic ratio was (27.4 ±1.8)%, (49.7 ±1.4)%, (65.1 ±1.4)%, and (69.0 ±2.0)%, respectively (F= 1112.767, P< 0.01). The induction of apoptosis showed time and dose dependence (both P< 0.01). There was a trendency that the apoptotic ratio of SGC-7901 cell line was higher than that of the MKN-45 cell line at the same condition. RT-PCR showed that mRNA relative expression level of MMP-2 was 0.64±0.02, 0.49±0.01, 0.36±0.02, and 0.32±0.01, respectively (F= 274.321, P< 0.01) of negative control group and 5, 10, and 20 mg/ml trametes extract group after the effect of trametes extract on gastric cancer MKN-45 cell line. The relative expression level of MMP-9 in MKN-45 cell line was 0.71±0.01, 0.54±0.02, 0.47±0.02, and 0.39±0.02, respectively (F=203.948, P< 0.01). The expression level of MMP-2 in SGC-7901 cell line was 0.64±0.01, 0.42±0.02, 0.34± 0.20, and 0.29±0.01, respectively (F= 305.877, P< 0.01), while the mRNA expression level of MMP-9 was 0.65 ±0.15, 0.47 ±0.01, 0.44 ±0.01, and 0.39 ±0.02, respectively (F= 265.259, P< 0.01). Compared with the negative control group, the expression levels of MMP-2 and MMP-9 of the two cell lines were both decreased, after incubated with trametes robiniophila at the different concentrations (all P< 0.05), and with the addition of concentration, the expression levels of MMP-2 and MMP-9 were decreased. Conclusion Trametes robiniophila can induce the apoptosis of human gastric carcinoma cell lines MKN-45 and SGC-7901 in vitro and can inhibit the expressions of MMP-2 and MMP-9 and the effect of trametes robiniophila may be related with the differentiation degree.

ABSTRACT

Objective To investigate the effect of minimally invasive surgery in patients with midge-upper advanced gastric cancer (AGC). Methods 167 patients with middle-upper AGC underwent total gastrectomy+D2 lymph node dissection from January 2009 to December 2013 were enrolled in the study and divided into laparoscopic assisted total gastrectomy (LATG) group (n = 57) and open total gastrectomy (OTG) group (n = 110). Baseline data, operative, postoperative data and follow-up result were compared between the two groups. Results There were no significant differences in baseline data between the two groups (P > 0.05); The intraoperative blood loss and incision length in LATG group were significantly lower than that in OTG group (P 0.05); Postoperative first aerofluxus time, ecovery liquid diet time, postoperative hospitalization days and VAS score of pain in 3 d after operation in LATG group were significantly lower than that in OTG group (P 0.05); There was no patient dying within 30 d after surgery.The median follow-up time was 38.2 months (1~58 months). Postoperative 1, 3 year overall survival rates in LATG group and OTG group were 87.7% and 83.6%, 71.9% and 64.5%, and there were no significant differences between the two groups (P > 0.05). Conclusion For patients with middle-upper AGC, LATG+D2 lymph node dissection has lots of advantages including less intraoperative blood loss, shorter incision, faster postoperative recovery, shorter hospital stay and so on, which is safe and effective, and their short-and long-term results are satisfactory.

ABSTRACT

Recent studies showed that gastric mucosa was more susceptible to injury by invasion factors with aging, however,the studies were mainly on gastric antral mucosa,fundic mucosa was rarely studied.Aims:To investigate the effect of aging on related biological activity factors in gastric fundic mucosa in beagle dogs.Methods:Nineteen beagle dogs were assigned into younger group (aged 1-5 years),junior elderly group (aged 6-8 years)and senior elderly group (aged≥9 years).The contents of MDA,LPO,MPO in gastric fundic mucosa were determined by TBA method.The contents of PTEN,TE,survivin,caspase-3,caspase-9,ZO-1,CGRP,VEGF,COX-1 and COX-2 were assessed by ELISA.Results:Compared with younger group and junior elderly group,contents of MDA,LPO,MPO,PTEN,TE,ZO-1 ,CGRP,VEGF, COX-1 and COX-2 were significantly increased in senior elderly group (P0.05 ).The content of survivin in junior elderly group and senior elderly group was significantly decreased when compared with younger group (P =0.000 ).Conclusions:Disadvantaging changes of biological activity factors are found in gastric fundic mucosa in elderly beagles dogs,however, gastric mucosal blood flow, mucosa regeneration and epithelial tight junction related biological activity factors are significantly increased in senior elderly beagle dogs,which may be a phenomenon of degeneration-compensation.

ABSTRACT

Objective To investigate the effect of alizarin on BKCa channel beta subunit and electrophysiological characteristics in interlobar renal artery of SHR.Methods Tail artery pressure measurement instrument,pressure myograph system,whole cell patch clamp technique and Western blot were used to measure the change of systolic pressure of SHR,renal interlobar arterial diameter,single vascular smooth muscle cell electrophysiological characteristics and expression of the BKCa channel beta subunit before and after alizarin intervention,respectively.Results (1) SHR systolic pressure was decreased after alizarin intervention (P < 0.01).(2) ibTX inhibited alizarin-mediated renal interlobar arterial relaxation (P < 0.01).(3) Alizarin enhanced BKCa channel-mediated outward currents (P < 0.05).(4) Alizarin up-regulated BKCa channel beta subunits after alizarin intervention (P < 0.01).Conclusion Alizarin reduced SHR systolic pressure and relaxed interlobar renal artery by enhancement BKCa currents via mediation of the function of beta subunits.

Background:It has been demonstrated that H + -K + -ATPase expression in human parietal cells had a tendency to increase with aging. However,the effect of aging on activity of H + -K + -ATPase is still unclear. Aims:To investigate effect of aging on activity of H + -K + -ATPase. Methods:Fifteen male Sprague-Dawley(SD)rats were divided into 4,21,24, 27 and 30 months group,and 19 healthy beagle dogs were divided into younger group,junior elderly group and senior elderly group. The activity of H + -K + -ATPase in gastric fundal mucosa was assessed. Results:The activity of H + -K + -ATPase in gastric fundal mucosa in 4,21,24,27 and 30 months old rats were(4. 850 ± 0. 312)μmol·mg - 1 ·h - 1 , (5. 466 ± 0. 379)μmol·mg - 1 ·h - 1 ,(6. 068 ± 0. 228)μmol·mg - 1 ·h - 1 ,(5. 733 ± 0. 767)μmol·mg - 1 ·h - 1 and (6. 223 ± 0. 428)μmol · mg - 1 · h - 1 ,respectively. With aging,H + -K + -ATPase activity in rats had a tendency to increase(F = 4. 519,P = 0. 031). The activity of H + -K + -ATPase in beagle dogs in younger group,junior elderly group and senior elderly group were(11. 087 ± 4. 320)μmol·mg - 1 ·h - 1 ,(8. 549 ± 3. 250)μmol·mg - 1 ·h - 1 ,(12. 071 ± 2. 820)μmol·mg - 1 ·h - 1 ,respectively. There was no statistically significant difference among the three groups(F =1. 339,P = 0. 290). Conclusions:With aging,the activity of H + -K + -ATPase in rats and beagle dogs does not decline, but even has a tendency to increase.

ABSTRACT

Objective To evaluate the value of fecal high mobility group box-1 (HMGB 1 ) in early diagnosis and prediction of the severity of neonatal necrotizing enterocolitis (NEC). Methods From July 2013 to June 2015 , the neonates who had distention, vomit, or gross blood in stool and were suspected of NEC were recruited as NEC group while hospitalized children without abdominal distension, vomiting, bloody diarrhea, or other gastrointestinal symptoms were recruited as the control group. Stool samples were collected on day 1 , 3 , 5 and 7 after admission. The level of HMGB 1 was measured by enzyme linked immunosorbent assay (ELISA). Results In the end, there were 46 cases in NEC group and 15 cases in control group. In NEC group, 29 cases were conifrmed of stageⅠof NEC by abdominal radiograph within 24 h after hospitalized, all of them were deteriorated to stageⅡphase in 4 days, and 10 cases were deteriorated to stageⅢ. Seventeen cases were conifrmed of stageⅡby abdominal radiograph within 24 h after hospitalized, 7 cases were deteriorated to stageⅢ. In 17 stageⅢcases, 11 cases received surgical treatment and 6 cases gave up. Eight cases survived and 3 died after surgery. The levels of HMGB 1 in NEC group on day 1 , 3 , 5 and 7 after hospitalized were higher than those in control group (P

ABSTRACT

Objective To investigate the effect of aging on gastric pepsinogen secretion through observing histological changes of gastric fundal mucosa and ultrastructure of gastric chief cells in human and Beagle dogs at different ages.Methods Fifty middle-aged and elderly indigestion patients with gastroscopy were selected as study objects and divided into young and middle age group (age 20-59 years,n=19),junior elderly group (age 60-69 years,n=11),middle elderly group (age 70-79 years,n=10),and senior elderly group (no less than 80 years,n=10).In addition,nineteen healthy Beagle dogs were also selected as study objects and divided into young and middle age group (age between 1 to 5 years,n=8),junior elderly group (age six to eight years,n=5),and senior elderly group (no less than 9 years,n=6).The histology and morphology of gastric fundal mucosa of human and Beagle dogs were observed under light microscope.Then the thickness of lamina propria was measured and the number of gastric chief cells was counted.The ultrastructure of gastric chief cells was observed under electron microscope,and the area percentages of secretory granule(also called mucous granule)area in cytoplasm of chief cell was calculated.The data was analyzed by one-way analysis of variance.Results No significant histological and morphological changes of gastric fundal mucosa were observed in human and Beagle dogs at different ages.There was no significant difference in the thickness of lamina propria among different ages in human and Beagle dogs (all P>0.05).The number of human chief cells of young and middle age group,junior elderly group,middle elderly group and senior elderly group was 71.79±16.85,52.73±16.60,57.10±20.21 and 43.70 ±16.89/high power field (HPF),respectively,which decreased as age increased (F=6.431,P=0.001).The numbers of chief cells of young and middle age group,junior elderly group,and senior elderly group of Beagle dogs were 328.38 ±32.36,341.20 ±42.49 and 225.67 ± 52.19/HPF,respectively,which decreased as age increased (F=13.647,P<0.01).Aging-related degeneration was founded in ultrastructure of gastric chief cells in human and Beagle dogs including rough endoplasmic reticulum dilation,ribosones missing,secretory granule decreasing and so on.The area percentages of secretory granule area in cytoplasm of human chief cells of young and middle age group,junior elderly group,middle elderly group and senior elderly group were (67.28±3.79)%,(66.88±4.84)%,(65.63±7.10)% and (56.25±8.47)%,respectively,which decreased as age increased (F=6.069,P=0.002).The area percentages of secretory granule area in cytoplasm of Beagle dogs' chief cells of young and middle age group,junior elderly group,and senior elderly group were (58.83 ± 3.07) %,(52.34 ± 4.73) % and (39.89 ± 4.46) %,respectively,which decreased as age increased (F=38.837,P<0.01).Condusion As age increased,the ability of pepsinogen secretion of fundal gland decreased,which may be one of the reasons of functional dyspepsia in the eldly.

ABSTRACT

Background:It has been demonstrated that acid secretion-related gastric fundic structures do not degenerate in elderly individuals. As specimens obtained from human body are greatly influenced by environmental factors,the results need to be further verified by standard animals. Aims:To investigate the aging-related changes of histology of gastric fundus and ultrastructure of fundic gland in beagle dogs. Methods:Nineteen healthy beagle dogs were divided into three age groups,8 in younger group(aged 1-5 years),5 in junior elderly group(aged 6-8 years)and 6 in senior elderly group (aged ≥9 years). Gastric specimens obtained from the fundus were examined by light microscope,the thickness of lamina propria and percentage of parenchymal area of fundic gland in lamina propria were measured,and the parietal cells,chief cells and endocrine cells were counted. Ultrastructure of fundic gland cells were observed by electron microscopy,and the area percentages of related organelles were calculated. Results:The thickness of lamina propria in gastric fundus of beagle dogs showed a decline tendency with aging,but no significant differences were found among the three age groups(P >0. 05);in senior elderly group,a marked decrease in density of fundic gland was observed and with an increase of interstitial tissues. The amount of parietal cells and the area percentages of mitochondria and tubulovesicle in parietal cells showed no aging-related reduction(P > 0. 05),while the amount of chief cells,area percentages of zymogen granule in chief cells,and mucous granule in mucous neck cells were significantly decreased in senior elderly group(P < 0. 05). The amount of endocrine cells increased gradually with aging(P < 0. 05). Conclusions:Aging-related degeneration is not existed in acid secretion-related parietal cells and organelles in gastric fundus of beagle dogs. Furthermore,endocrine cells which secret histamine,gastrin,and somatostatin,etc. for stimulating acid secretion even increased. These changes might be a compensatory mechanism for acid secretion in elderly individuals.

ABSTRACT

Objective To investigate the changes of liver histology and ultrastructure in aged Beagles dogs.Methods A total of fifteen healthy Beagles,1 .5 to 10.6 years old,were divided into middle-aged group (n=8,average (2.21 ±0.94)years old)and aged group (n=7,average (8.26±1 .52) years old)according to age.After intravenous anesthesia,the liver tissues of dogs were taken to compare number and morphology of hepatocytes and extra cellular matrix (ECM)of two groups under light microscope with hematoxylin-eosin staining.The number density, morphology and area fraction of mitochondria,the morphology and area fraction of endoplasmic reticulum,the changes of number density and morphology of microbody,the thickness of hepatic sinusoidal endothelial cell as well as the density and the diameter of fenestrae,the changes of morphology and number of lipofuscin in two groups were compared under electron microscope.Independent sample t-test was performed for comparison between two groups.Results Under light microscope,hepatocytes were arranged radially around the central veins,especially obvious in the middle aged group. Hepatocytes were circular or oval,most were mononuclear or binuclear.The cytoplasma of hepatocyte in middle-aged group was dense eosinophilic pink.Vacuolar lipid droplets were small and scanty.The cell boundary was clear.In aged group, cytoplasmic vacuolization of hepatocyte was more obvious,bigger vacuolated lipid droplets were observed, eosinophillic granules were scattered in the cytoplasma,cellular boundary was blurry and the number of hepatocytes significantly decreased when compared with middle-aged group ((75 .80 ±5 .76 )/high power field (HPF)vs (97.80 ±6.45 )/HPF,t = -2.510,P =0.026 ).There were no obvious infiltration of inflammatory cells in the liver tissue of Beagles dogs in both middle-aged group and aged group.Compared with middle aged group, cell gap and ECM increased in aged group. Under electron microscope, mitochondria of normal hepatocyte was oval,round or horseshoe-shaped with double membranes,ridge was observed inside and distributed around nucleus.Compared with middle-aged group,the shape of hepatocyte mitochondria in aged group was varied,the number density of mitochondria significantly decreased (0.202±0.021 vs 0.248 ±0.040,t = -2.683,P =0.020)and the area fraction remarkably reduced (0.136 ±0.015 vs 0.202 ±0.019,t = -6.083,P <0.01 ).Endoplasmic reticulum was mesh laminated arranged,scattered flake distributed and was mostly seen around nucleus in flat or stacked shape.Compared with middle aged group,the area fraction of endoplasmic reticulum of hepatocyte in aged group was obviously reduced (0.006±0.001 vs 0.011 ±0.004,t=-3.338,P =0.005).Lipofuscin was shaped as round or irregular structure without membrane and pigment unevenly distributed inside.The number of lipofuscin in aged group was significantly higher than that in middle-aged group (0.028±0.025 vs 0.001 ±0.000,t=2.880,P =0.014).The number of microbody in aged group was slightly less than that of middle aged group and the difference was not statistically significant (P =0.603).The thickness of the liver sinusoidal endothelium in aged group significantly increased compared with that in middle-aged group ((0.242±0.047)μm vs (0.114±0.075)μm,t=3.881 ,P =0.002).The diameter and density of endothelial fenestration in aged group ((1 .895 ± 0.439 )μm and (0.260 ± 0.120 )/μm)obviously decreased compared with those in middle-aged group ((3.186±0.332)μm and (0.723±0.184)/μm,t=6.345 and -4.511 ,both P <0.01).Conclusions There were significant degradations of liver in quality and quantity among aged individuals.These degradations were the structure basis of decreased hepatic reserve function and involved directly or indirectly in high prevelance of hepatic diseases among elders.

ABSTRACT

Objective To analysis the malignant performance characteristics of tumor stem cell-like side popula-tion cells in patients with cervical cancer. Methods The cervical cancer cells were obtained from surgical resection tumor tissue. The tumor stem cell-like side population cells were isolated by flow cytometry. The cell growth curve was drawn by MTT assay. The invasion ability of tumor cells was compared by transwell assay. The clonogenic capacity was detected by clone formation in soft agar. The expression level of ABCG2 protein, a drug-resistant gene, was detected by immunofluores-cence method. Finally, these cells were transplated into the subcutaneous of de thymus mice. The rate of tumor formation was compared between groups. Results The results from flow cytometry assay showed the percentage of cervical cancer stem cell-like side population cells was 1.39%. Compared with the non-side population cells, the side population cells grow quickly, showed the enhanced invasion ability and colony forming ability. There was more high expression level in ABCG 2 protein of side population cells. The tumor form rate was 100%(10/10) in the side population cells and the non-side popula-tion cells was 20%(2/10). Conclusion The cervical cancer stem cell-like side population cells have more malignant perfor-mance characteristics than that of non-side population cells, which maybe a core target for cancer gene therapy in the future.

During the process of normal aging,digestive tract experiences a series of degenerative changes,which is the foundation of many gastrointestinal motility disorders. Further study on gastrointestinal dysmotility and its mechanisms is of great significance for the diagnosis and treatment of related diseases in aging population. In this article,the advances in studies on effects of aging on motility of digestive tract( including oropharynx,esophagus,stomach,small bowel,colon and anorectum)and its mechanism researches were reviewed.

ABSTRACT

Objective To investigate the isokinetic strength of knee extensors and flexors in patients with knee osteoarthritis (KOA), and to establish the correlation between the isokinetic strength and function in patients with KOA. Methods 23 patients with bilateral KOA and 14 matched normal controls finished the isokinetic test of knee extensors and flexors, the Five Times Sit-to-Stand Test (FTSST), Gait analy-sis, and Balance test. The KOA patients were evaluated with Visual Analog Scale (VAS) for pain and Western Ontario and McMaster Univer-sity Osteoarthritis Index (WOMAC). Results It was less of the peak torque, average peak torque, average power, max rep total work, total work of knee extensors and flexors in the mainly involved limbs than the contralateral limbs (P0.05). Conclusion It is weak of the isokinetic strength of knee extensors and flexors in patients with KOA in the mainly involved limb, as well as the isokinetic strength of knee extensors compared with the normal controls. The changes in the ex-tensors and flexors are not equivalent. The peak torque of knee extensors significantly correlated with the pain and functions of the knee.

Objective:To find out the impacting factors in uncertainty by analyzing the measurement uncertainty in the determina-tion of irbesartan tablets by HPLC and provide the evidence for the measurement evaluation. Methods:A mathematic model for calcu-lating uncertainty was established, and every component in the uncertainty was assessed. Results:The expanded standard uncertainty for the HPLC determination was 2. 4%. Conclusion:The measurement uncertainty of the experiment is mainly caused by the measur-ing method.

ABSTRACT

Objective To investigate the relationship between the efficacy of autologous cultured melanocyte transplantation and serum levels of interleukin-17 (IL-17) and FoxP3 in patients with vitiligo.Methods Forty patients with stable vitiligo vulgaris were included in this study,and received autologous cultured melanocyte transplantation.Six months after the transplantation,treatment efficacy was evaluated,and patients were classified into the successfully treated group (n =25) and unsuccessfully treated group (n =15).Peripheral blood was collected from all the patients before the transplantation,and enzyme-linked immunosorbent assay (ELISA) was performed to measure the serum levels of IL-17 and FoxP3.Statistical analysis was done using two independent samples t-test with the SPSS software (version 17.0).Results The successfully treated patients showed lower serum levels of IL-17 ((15.29 ± 7.86) vs.(43.88 ± 13.02) ng/L,P < 0.05),but higher serum levels of FoxP3 ((6.08 ± 2.03) vs.(3.37 ± 1.81) ng/L,P < 0.05) than the unsuccessfully treated patients.Conclusion The increased serum IL-17 and decreased serum FoxP3 may contribute to the failure of autologous cultured melanocyte transplantation in patients with vitiligo.

Gastrointestinal tumors are the major disease that threaten human health.According to statistics,more than 700 000 people died every year because of gastric cancer,1/10 of all cancer mortality.Colorectal cancer incidence of malignant tumors,which ranked third,ranked fourth in cancer deaths.At now the treatment of gastrointestinal tunors mostly are operation,radiotherapy and chemnotherapy,but to spread of tumors,doctors still haven' t more better method.Nanoparticles have properties in the diagnosis and treatment of cancer,including their size-dependent properties,stability in solvent,ideal size for delivery within the body,and tunable surface chemistry for targeted delivery.Here,the author do an overview of nanoparticles in gastrointestinal tumors ablation.

ABSTRACT

Objective To determine the role of APPL1,an adaptor protein,played in pancreatic β-cell.Methods APPL1 was overexpressed in INS-1 cells with adenovirus encoding APPL1.Western blot was conducted to measure protein cxprcssion.Propidium iodide/Hoechst staining was used to determine the cell apoptosis.Insulin secretion was measured by ELISA.Results Exposure of INS-1 cells to 20 mmol/L glucose or 30U/ml interleukin-1 β plus 20 ng/ml TNF-α 48 h induced β-cell apoptosis (P<0.01) and impaired 2 h glucosestimulated insulin secretion (P< 0.01).Overexpression of APPL1 in INS-1 decreased cell apoptosis by 34.16%-42.79% (P<0.01) and increased glucose-induced insulin secretion by 1.39-2.20 folds compared with control groups (P<0.05).Conclusion APPL1 decreases β-cell apoptosis and increases glucose-stimulated insulin secretion,and thus protects β-cell against high glucose or cytokines-induced dysfunction.

ABSTRACT

Tumor invasion and metastasis are regarded as main reasons for the failure of therpy and the reason of patients death.The mechnism of tumor metastasis is still uncertain.The pre-metastatic niche hypothesis provides us with new ideas to discover the mechnism.Numerous materials are involved in the formation of the pre-metastatic niche according to this hypothesis,including bone marrow-derived cells,microvesicles,exosomes,CD44,and so on.A further research on this hypothesis helps to deeply understand the nature of metastasis and leads clinical doctors to explore novel targets for clinical diagnoses and therapies.
